The Biochemical Properties of NAD+ Analogs: NGD and NHD

For instance, they have shown some affinity for enzymes like Alcohol Dehydrogenase (ADH) in vitro, although with significantly lower affinity compared to NAD+. This indicates a potential, albeit weaker, capacity to participate in electron transfer reactions that are fundamental to cellular metabolism.
Crucially, current research suggests that NGD and NHD do not function as substrates for key NAD+-dependent enzymes like Sirtuins and Poly(ADP-ribose) polymerases (PARPs). This distinct biochemical property differentiates them from NAD+ and implies that their impact on cellular processes like DNA repair or deacetylation may be limited or absent. Further research is needed to clarify whether specific enzymes exist that preferentially utilize these analogs, or if they act as modulators of NAD+ metabolism itself.
